Technical Questions

What is CHD gene-based sexing?

CHD (Chromobox Helicase DNA-binding) genes are located on the avian sex chromosomes (Z and W). In most bird species, the CHD-Z gene is present on the Z chromosome and the CHD-W gene is unique to the W chromosome (female-specific). PCR amplification using conserved primers (such as P2/P8) produces:

  • Male (ZZ): Single band (CHD-Z, ~400-650 bp depending on species)
  • Female (ZW): Double bands (CHD-Z + CHD-W, ~350-650 bp depending on species)

This method works across most bird species because the CHD genes are highly conserved in avian evolution.
