Swab Collection Protocol (Disease Testing)
Swab samples are the preferred sample type for avian disease testing at SENO. Oral and cloacal swabs are collected using sterile flocked swabs and immediately placed into nucleic acid stabilization buffer to preserve pathogen RNA and DNA for downstream molecular detection.
When to Use Swab Samples
Swab-based disease testing is indicated in the following scenarios:
- Respiratory disease investigation — Oral/choanal swabs for detection of respiratory viruses and bacteria
- Enteric disease investigation — Cloacal swabs for gastrointestinal pathogen screening
- Comprehensive health screening — Combined oral + cloacal swabs for full pathogen panel
- Quarantine or import screening — Pre-movement health clearance testing
- Outbreak investigation — Rapid collection from multiple birds during disease outbreaks
- Neonatal testing — Safer than blood collection in very young chicks

Oral Swab Procedure
Preparation
- Put on a fresh pair of sterile gloves
- Remove a sterile flocked swab from its packaging (avoid touching the tip)
- Open the stabilization buffer tube and set the cap aside (keep clean)
Collection Steps
- Gently restrain the bird — towel restraint is recommended for parrots and other psittacines
- Open the beak by applying gentle pressure on the commissures (corners of the mouth)
- Insert the swab into the oropharynx — the cavity at the back of the mouth
- Rotate the swab against the mucosal surface for 5–10 seconds with gentle pressure
- Swab the choanal slit — the narrow opening on the roof of the mouth (palate)
- Rotate again for 3–5 seconds against the choanal mucosa
- Withdraw the swab carefully — avoid touching the beak, tongue, or outside of the mouth
- Place the swab tip into the stabilization buffer tube
- Break or cut the swab handle at the break line (most flocked swabs have a pre-scored break point)
- Seal the tube tightly — ensure the cap is fully closed to prevent leakage

Cloacal Swab Procedure
Preparation
- Use a fresh pair of sterile gloves (even if collecting oral and cloacal from the same bird)
- Remove a new sterile flocked swab from packaging
- Open a separate stabilization buffer tube
Collection Steps
- Restrain the bird with the vent (cloacal opening) exposed
- Moisten the swab tip with sterile saline or stabilization buffer (optional — reduces friction)
- Insert the swab approximately 0.5–1 cm into the cloaca (do not force)
- Rotate the swab gently for 3–5 seconds — the swab tip should contact the cloacal mucosa
- Withdraw the swab and inspect the tip for visible fecal material (a small amount is expected)
- Place the swab tip into the stabilization buffer tube
- Break the swab handle at the break line and seal the tube

Combined Oral + Cloacal Swab Protocol
For comprehensive disease screening, SENO recommends collecting both an oral and a cloacal swab from each bird. These can be placed into a single stabilization buffer tube (one swab per tube is still acceptable). When combining:
- Collect the oral swab first (less contaminated)
- Place the oral swab into the stabilization tube and break the handle
- Collect the cloacal swab with a fresh swab
- Add the cloacal swab to the same tube
- Seal the tube and label as "Combined Oral + Cloacal"
Stabilization Buffer Handling
The nucleic acid stabilization solution provided by SENO contains chemical agents that:
- Inactivate cellular and microbial nucleases (RNases and DNases)
- Precipitate nucleic acids in a protected state
- Prevent microbial growth during transport
- Maintain DNA and RNA integrity at ambient temperatures for up to 14 days

WARNING: The stabilization buffer contains guanidine salts. Avoid contact with skin and eyes. If contact occurs, rinse with copious amounts of water. Do not ingest. Do not mix with bleach or chlorine-containing disinfectants — toxic gas may be produced.

Labeling
Label each tube immediately after collection with the following information using a waterproof marker:
- Bird ID (or band number)
- Collection date (YYYY-MM-DD format preferred)
- Sample type (Oral / Cloacal / Combined)
- Collector initials

Packaging for Shipment
- Place sealed tubes upright in a tube rack or padded container
- Place the rack inside a sealed biohazard bag
- Place the biohazard bag inside an insulated box
- Add cold packs (conditioned at 4°C) for ambient-temperature shipments
- Add dry ice for frozen shipments — declare as UN3373 Biological Substance, Category B
- Include the SENO submission form in a waterproof sleeve on top of the cold pack (not frozen)
Quality Control Checkpoints
Before sealing the shipment, verify:
