Passerines (Canaries, Finches & Songbirds)¶
Overview¶
SENO tests a variety of passerine species (songbirds), primarily for DNA sexing applications. Many passerines are sexually monomorphic — males and females share near-identical plumage — making DNA testing the only reliable method for gender identification outside the breeding season when song distinguishes males.
Background¶
The passerine order (Passeriformes) is the largest order of birds, containing over 5,000 species — more than half of all bird species. Fringillidae (finches) and Estrildidae (waxbills) are the most commonly tested families, driven by the aviculture and exhibition communities.
Passerines present unique challenges for molecular testing:
- Small body size: Canaries weigh 15–30 g, finches as little as 10–20 g — limiting blood collection volume
- Small feather follicles: A canary feather follicle yields 5–15 ng DNA vs. 50–100 ng for a macaw
- Fragile nature: Stress from handling during sample collection can be dangerous for very small birds
- CHD fragment resolution: Some species produce CHD-W and CHD-Z fragments that are close in size
- Disease profile: Different from psittacines — PBFD is not a concern in passerines, but APV and AIV can be

Visual sexing accuracy is 0% for juvenile birds of all species — making DNA testing the only option for nestlings and young birds.
Collection Tips for Small Passerines¶
Feather Collection (Preferred Method)¶
- Use 5–7 feathers minimum (small follicle = less DNA)
- Pluck from the chest/breast region — follicles here are richest in DNA
- Hold the bird in a darkened cloth/Towel (less stressful than open handling)
- Support the bird's body — pressure on the chest can restrict breathing in birds < 20 g
- Do not pluck tail feathers: they contain less DNA and take longer to regenerate
- Inspect each feather base: the follicle should appear white and bulbous; discard if dry
Blood Collection¶
- Maximum 0.05 mL (50 µL) for birds under 15 g; 0.1 mL for birds up to 30 g
- Use a 27–30G needle for venipuncture (very fine)
- FTA card blood spots preferred — less stress than EDTA tube collection
- Apply gentle pressure to puncture site for 10–15 seconds after collection to prevent hematoma
- A single drop of blood (10–20 µL) on an FTA card is sufficient for both sexing and basic disease PCR

Most passerine species tested at SENO show a ~15 bp difference between CHD-W and CHD-Z with the P2/P8 primer pair, which is reliably resolvable on a 3% agarose gel or capillary electrophoresis system. The 2550F/2718R primer set serves as backup if co-migration is suspected.

Frequently Asked Questions¶
Q: Can I sex a canary by its colour? A: No. Canary colour (yellow, white, red, variegated) is determined by autosomal genes that are not sex-linked. Red Factor canaries are not all male — females also express the red pigment. Colour is a completely unreliable indicator of sex in canaries.
Q: Why is the failure rate higher for finch samples than for parrot samples? A: The primary reason is DNA quantity. A canary feather follicle may contain 10× less DNA than a macaw feather follicle. If the feather is not freshly plucked with an intact follicle, or if an insufficient number is submitted, the extracted DNA may fall below the threshold needed for reliable PCR amplification.
Q: How can I visually estimate the sex of my Gouldian Finch? A: Adult Gouldian Finch males have a brighter purple chest, deeper green back, and more vivid yellow belly than females. However, color intensity varies by individual, and birds less than 6 months old cannot be sexed visually at all. Young Gouldian finches of both sexes look identical. DNA testing is the only reliable method for juveniles.
Q: Do male canaries sing? A: Yes — sustained, complex song is a male characteristic. Female canaries may chirp and make brief calls but do not produce the elaborate song of the male. However, juvenile males don't sing (typically start at 6–10 months), and some males sing only during the breeding season. A non-singing bird cannot be assumed to be female.
Q: Can I test my finch for APV? A: Yes. Avian Polyomavirus can infect finches and canaries, though it is less common than in budgerigars. Testing is recommended if you have unexplained deaths in juvenile finches or are introducing new birds to an existing collection.
Q: Is it safe to pluck feathers from a Gouldian Finch? A: Yes, when done correctly. Gouldian Finches are delicate (14–17 g) but tolerate chest feather plucking if the procedure is quick and the bird is handled gently. Use a soft cloth to hold the bird, pluck only 5–7 chest feathers, and return the bird to its cage immediately. Do not hold for more than 30 seconds. For very nervous birds, consider FTA card blood spot collection instead (requires single drop of blood).
