Background

Psittacines (order Psittaciformes) encompass over 350 species of parrots, macaws, cockatoos, lorikeets, lovebirds, and budgerigars. They represent the largest group of captive birds tested at SENO, accounting for approximately 55% of all DNA sexing samples and 80% of disease testing samples. Parrots are prominently represented because: (1) many species are sexually monomorphic and require DNA sexing, (2) they are long-lived (20–80+ years) and often kept as companion animals, (3) they are susceptible to several species-specific viral diseases, and (4) the international pet trade and aviculture community demands reliable testing.

DNA Sexing Considerations

Small Psittacines (Lovebirds, Budgies, Parrotlets)

  • Use 5–7 feathers for DNA extraction (small follicles = limited DNA yield)
  • CHD-W and CHD-Z fragments with P2/P8 may be only 5–10 bp apart
  • Capillary electrophoresis strongly recommended in place of gel-based analysis
  • Disease screening: combined oral/cloacal swab recommended
  • Lower blood volume limits in very small species (≤ 0.1 mL for birds under 50 g)

Medium Psittacines (Conures, Cockatiels, Ringnecks, Small Amazons)

  • Use 3–5 feathers sufficient for both sexing and disease testing
  • P2/P8 primer pair generally provides adequate separation
  • Gel electrophoresis (2.5–3% agarose) usually sufficient for fragment discrimination
  • Disease screening: separate oral swab for APV/PBFD if clinical signs present

Large Psittacines (Macaws, Cockatoos, African Greys)

  • 2–3 feathers sufficient for sexing — the follicles are large and DNA-rich
  • Preferred blood volume: 0.3–0.5 mL for disease testing
  • P2/P8 primer pair gives reliable separation (15–25 bp difference typically)
  • PBFD screening recommended for all new arrivals before introducing to collections
  • Regular (quarterly to semi-annual) PBFD testing recommended for multi-bird facilities
